It begins at a partial cloverleaf interchange with Interstate 27 at exit 33, where the route quickly descends underground into the Vanderbilt Tunnel. The tunnel is yet to be completed, however exit numbers have already been planned.
The entirety of the Vanderbilt Thruway is limited to passenger vehicles due to low clearance in the tunnel. Commercial vehicles are not permitted and must use alternate routes, like Colonia Route 31W.
